NovoTyr Therapeutics Ltd. (“NovoTyr”) was founded as a Meytav Incubator portfolio company on March 2005. The founder, Prof. Alexander Levitzki, is a leading researcher and instigator in the field of tyrosine kinases as targets for therapy, and was recently awarded the prestigious Wolf prize for his contribution in this field. He pioneered the concept and development of tyrosine phosphorylation inhibitors (tyrphostins) as a strategy to treat malignancies as well as non-cancer proliferative diseases and inflammatory conditions. The company has developed novel proprietary inhibitors with several unique features, showing high anti-cancer potential in-vitro and in-vivo. Sector and Field of Activity Pharmaceuticals, Biotechnology, Drug Development, Cancer and other proliferative diseases Underlying Science and Technology NovoTyr develops novel inhibitors of insulin-like growth factor receptor (IGF1R), as drugs for the treatment of cancer and other inflammatory disorders with unique properties and profile of activities. The ADVANTAGES of NovoTyr's product: * Unique approach & long-lasting effects: NovoTyr developed a unique IGF1R inhibitor, which inhibits the tyrosine kinase activity of IGF1R, but as opposed to other reported inhibitors, it also induces a long-lasting elimination of the protein levels of IGF1R direct substrate IRS1. Thereby, NovoTyr's inhibitor blocks IGF1R signaling for a very long period, leading to the cancer cell apoptosis (programmed cell death). * The inhibitors are proprietary novel small molecules. The ADVANTAGES of the target protein: * Specificity: In contrast to normal cells the viability of various cancer cells is dependent on IGF1R activity. Therefore, IGF1R inhibitors selectively affect tumor cells with little or no toxic effects on normal cells. * Target is expressed throughout the disease progression: IGF1R inhibitors are developed to treat primary tumors as well as advanced metastatic cancers. * Synergistic effect with other modalities: IGF1R inhibitors will be used as a mono-therapy or as chemosensitizers, when combined with conventional chemotherapy or radiation. The latter strategy is based on the observation that IGF1R activity is enhanced during the development of resistance of cancer cells to chemotherapy, and its inhibition sensitized these cells to apoptosis (programmed cell death). UPDATES: During its first year of activity NovoTyr developed a novel and potent inhibitor of IGF1R, named NT52, with several unique features: NT52 induces a specific reduction in the levels of IGF1R-direct substrate IRS1 in cancer cells. This effect is long-lasting and correlates with high anti-cancer activity in-vitro and in-vivo. NT52 leads to the inhibition of the central anti-apoptotic pathway in cancer cells, the activation of PKB, and to a subsequent apoptosis of these cells. In a successful process of lead-optimization, NovoTyr designed and characterized improved 2nd generation proprietary leads, covered by a recently filed patent application. In-vivo experiments, evaluating the efficacy of NT52 and its improved derivatives in human tumor-transplanted nude mice, show significant and pronounced inhibitory effects in prostate cancer, ovarian cancer and melanoma models. NovoTyr will initiate preclinical development of the selected lead compound at 2009. In 2008, Teva Pharmaceutical Industries (Nasdaq: Teva) signed an investment agreement in the Novotyr. Teva and Novotyr will collaborate in the development of these novel inhibitors, and following phase II clinical studies, Teva will have the option to a worldwide and exclusive license for further development and marketing of the drug.
